Anti-ageing ingredients that actually work: the 4 backed by science

Quick answer

Out of the endless list of ingredients promising to knock years off your face, only four have solid evidence behind them: retinol (the most studied anti-wrinkle active there is: it speeds up cell turnover and stimulates collagen), peptides (messengers that tell your skin to build new collagen), antioxidants (vitamin C, vitamin E, ferulic acid, carotenoids: they neutralise the free radicals that break down the collagen you still have) and sunscreen (the only one that tackles the number-one cause of skin ageing). Everything else — snail mucin, salmon sperm, colloidal gold and friends — may feel lovely, but right now it doesn't have the backing to promise you fewer wrinkles. If you're only going to add two things to your routine: retinol at night and sunscreen during the day. The rest is fine-tuning.

Snail mucin, dragon's blood, salmon sperm, whale placenta… it seems those of us in the cream business have a real weakness for shiny new ingredients that promise to be 'x1000000 more powerful' than the last one… and, as you've probably guessed by now, young padawan, NOT everything works.

So we've done the dirty work: separating the anti-ageing ingredients with actual studies behind them from the ones with nothing but a good marketing team. Spoiler: the good list is short and pretty boring. Which is exactly what you want when you're dropping €50 on a face cream.

What does it actually mean for an anti-ageing ingredient to work?

Sounds obvious. It isn't. For the purposes of this article, 'it works' means 1) it reduces wrinkles and 2) that can be measured objectively. Simple enough, right? Let's get into it:

It reduces wrinkles

Wrinkles are 'folds' that appear in the skin as collagen and elastin drop off in the upper layers — two proteins that, like bricks and mortar in a wall, keep your skin firm and taut. When we're 'young' (up to about 20) our skin's factory churns these proteins out at full speed, but after that peak the factory slows down and 'loses productivity' at a rate of roughly 1% a year. Any ingredient that gets the factory working again… will end up reducing your wrinkles.

It can be measured

We don't want to get too technical. But measuring 'properly' means an assessment by someone independent (if a brand 'pays'… for a study… you can't assume objectivity), with a control group so you can tell apart the people who used it from the people who didn't, repeatable without the results changing (more than one single study) and with a reasonably representative sample (results in a decent number of people, not just a handful of mates).

How to read a label: "clinically proven" sometimes means a double-blind trial with 60 people… and sometimes it means 22 volunteers filled in a questionnaire. If a study doesn't tell you how many people, how long for and compared to what, it's advertising, not evidence.

If we don't have those two things, does that mean the ingredient doesn't work? Neither yes nor no. Just that we need to keep gathering proof. That said, these are the 4 ingredients that meet both conditions (so far).

1. Retinol: the night-shift renovator

Retinol, a form of vitamin A, is known for its ability to stimulate cell turnover and boost collagen production. It helps reduce wrinkles and expression lines, improves texture and lends a hand with breakouts. It is without question the most studied ingredient in the category, going back to the 1950s. RETINOL — not derivatives, not double-barrelled versions (fewer studies, or less efficacy).

Translated: it does several things at once. It speeds up cell turnover, stimulates the synthesis of collagen and of your own hyaluronic acid, slows the enzymes that break collagen down, and regulates sebum. It's the only cosmetic active that works on wrinkles, texture, dark spots and breakouts at the same time.

What the studies say

This isn't brochure evidence. In a randomised, double-blind, vehicle-controlled trial at the University of Michigan (Kafi et al., 2007), topical retinol significantly reduced fine wrinkles and increased the production of collagen and of glycosaminoglycans — the ones that hold water — in volunteers with an average age of 87. So, never too late. A later comparative study (Kong et al., 2016) measured a significant reduction in wrinkles at 12 weeks, with histological and collagen gene expression changes similar to those of retinoic acid (the prescription drug), but far better tolerated. And a 12-month follow-up (Abed et al., 2023) points to 0.3% as the concentration that reorganises skin structure without wrecking your face.

The small print: retinol needs 8 to 12 weeks to show up on wrinkles, it's a night-only active, and it makes daily sunscreen non-negotiable. Anyone promising results in a week is selling you an Instagram filter.

2. Peptides: the firmness messengers

Peptides are short chains of amino acids that act as messengers in the skin, telling the 'builder cells' to produce your skin's 'construction materials'. There are many types, sizes and functions… and, to be honest, they have more evidence in vitro (on skin cultures grown in a lab) than in vivo (on actual people).

That doesn't disqualify them — it places them. A well-chosen peptide is a good complement to retinol, not a substitute for anything. And two families have made it into human trials.

What the studies say

Signal peptides — like Tripeptide-5 — mimic the natural activation of TGF-β, the pathway your skin uses to order new collagen (Schagen, 2017). Neurotransmitter-inhibiting peptides — the famous Acetyl Hexapeptide-8, or Argireline, the "botox-like peptide" — take a different route: they reduce the micro-contractions that form expression lines. A randomised, placebo-controlled trial measured a 48.9% reduction in total wrinkle severity at 4 weeks, and the most recent review (2025) confirms they reduce wrinkle depth and improve elasticity and hydration.

The small print: that same review notes that skin penetration of these peptides is still under study. Which is why you trust a formula with peptides and retinol, not one that only carries peptides and charges the same.

3. Antioxidants: the shields against ageing

Antioxidants protect the skin from the free radicals generated by external factors such as UV radiation and pollution. Those free radicals can damage skin cells and speed up ageing. Applying topical antioxidants neutralises them and prevents cell damage.

The difference with retinol is one of role, not category: retinol rebuilds what you've already lost; the antioxidant defends what you still have. That's why one goes on at night and the other during the day. The ones with the best paperwork: vitamin C, vitamin E, ferulic acid, niacinamide and plant-derived carotenoids — green algae (Dunaliella salina), goji berry or dragon's blood.

And here's the detail that separates a well-designed antioxidant serum from one that just has "vitamin C" on the label: antioxidants work as a team. Vitamin C covers the watery side of the cell, vitamin E the oily side, and ferulic acid stabilises both so they don't oxidise before they've done their job.

What the studies say

Healthy skin naturally contains very high concentrations of vitamin C, and the reference review on the subject (Pullar, Carr & Vissers, 2017) sums up its two established functions: stimulating collagen synthesis — it's an obligatory cofactor for the enzymes that stabilise it; no vitamin C, no collagen — and protecting against UV-induced photodamage. That same review is honest about the nuance almost no brand mentions: how much you gain from applying it topically versus simply getting enough in your diet is less well established than the marketing suggests.

The teamwork bit isn't just a nice phrase either: in the landmark study on this (Lin et al., 2005, Duke University), adding 0.5% ferulic acid to a solution of 15% vitamin C and 1% vitamin E improved the chemical stability of both vitamins and doubled the skin's photoprotection against solar-simulated radiation: from ×4 to ×8, measured by both erythema and sunburn cell formation.

The small print: the mechanism is solid; the visible result of an antioxidant on its own is modest. Its real job is to multiply your sunscreen, not replace it.

4. Sunscreen: the daily bodyguard

Sun exposure is THE main cause of premature skin ageing. UV rays damage collagen and elastin fibres, causing wrinkles and dark spots. Daily sunscreen use protects the skin from that damage.

There will come a day when sunscreen is like separating your recycling or not smoking indoors: first written into law… and then, a generation later, nobody can quite understand how we ever lived without it.

What the studies say

This is the ingredient with the most convincing evidence of the four, and with the hardest study design to pull off: a 4.5-year randomised trial in Australia (Hughes et al., 2013) compared people using sunscreen daily with people using it "when they remembered". Result: the daily-use group showed 24% less visible skin ageing. No anti-wrinkle cream on the market has a study that long.

The small print: even when it's cloudy, even if you never leave the office, and even if you tan easily. The sun that ages you isn't the beach sun: it's the walk-to-the-shops sun, over 30 years. If adding a step feels like too much, look for a moisturiser with high SPF in a single bottle.

And the ones that (still) don't: snail mucin and other wildlife

We're not saying they're scams or that they do nothing. We're saying something more boring and more honest: they don't yet meet the two conditions from the start of this article. If you like them, use them. Just don't ask them for what they can't give.

Snail mucin

Its mucin contains allantoin, glycoproteins and hyaluronic acid, and there are small studies showing results in hydration and texture. What's missing are large, repeated, independent trials measuring wrinkles. A good moisturiser; a poor substitute for retinol.

Salmon sperm (PDRN / polynucleotides)

There's genuinely interesting literature here… almost all of it injected in clinic, not applied in a cream. An active working with a needle doesn't mean it crosses your skin from a jar. That's exactly the leap marketing makes without telling you.

Colloidal gold, caviar, diamond and other jewellery

They raise the price and the luxury perception. There's no evidence they raise your collagen. If gold appears after the preservative on the INCI list, you know how much of it you're getting.

Plant stem cells

An apple stem cell doesn't "reprogram" your skin: what actually ends up in the cream are plant extracts and growth factors that, at best, behave like antioxidants. Brilliant name, modest mechanism.

Collagen in a cream

Collagen is a huge molecule. Applied on the surface it hydrates and forms a film, but it doesn't slip down into the dermis and become your collagen. That's what retinol, peptides and vitamin C are for — they tell your skin to make it itself.

How to build the routine in 3 moves

You don't need twelve steps or a wash bag the size of a carry-on. You need this:

  1. Night · Retinol. Clean, dry face and a cream with retinol at an effective concentration. Every other night for the first 2-3 weeks until your skin gets used to it; after that, every night. Peptides and soothing actives should already be inside the same formula.
  2. Day · Antioxidant + sunscreen. A serum or moisturiser with antioxidants and, on top, sunscreen. This step isn't optional: retinol leaves your skin more sensitive to the sun, and the sun is what gave you most of your wrinkles in the first place.
  3. Every month · Progress photo. Same light, same angle, relaxed face. The daily mirror is terrible at spotting slow change; two comparable photos two months apart are not.

Conclusion: choose your anti-ageing allies wisely

If what you're after is the ritual, the novelty or the nice packaging… buy the cream with whatever ingredients you fancy. Just don't ask it for miracles. To be sure your money is working, check that your routine includes retinol, peptides, antioxidants and sunscreen.

Which anti-ageing ingredients actually work?

Four: retinol, peptides, antioxidants (vitamin C, vitamin E, ferulic acid, niacinamide, carotenoids) and sunscreen. They're the only ones that meet both of our conditions: a measurable effect on wrinkles, and objective studies with a control group and a large enough sample to back it up.

What is the best anti-wrinkle ingredient?

Retinol, no contest. It's the cosmetic active with the most clinical trials behind it, going back to the 1950s: it speeds up cell turnover, stimulates the synthesis of collagen and of your own hyaluronic acid, slows the enzymes that break collagen down and regulates sebum. It's the only one that works on wrinkles, texture, dark spots and breakouts at once.

Do peptides really work, or is it marketing?

They work, with caveats. They have far more in vitro evidence (lab-grown skin cultures) than in vivo evidence (real people), and reviews note that their skin penetration is still being studied. Is vitamin C in a cream actually worth it?

Does snail mucin get rid of wrinkles?

There isn't enough evidence to say so. Its mucin provides allantoin, glycoproteins and hyaluronic acid, and there are small studies showing results in hydration and texture, but large, repeated, independent trials measuring wrinkle reduction are missing. As a moisturiser, fine; as an anti-ageing active, it doesn't compete with retinol.

What about salmon sperm (PDRN)?

The most interesting literature on polynucleotides and PDRN comes from treatments injected in clinic, not from topical cosmetics. An active working when delivered by needle doesn't mean it crosses the skin barrier from a jar of cream. Right now it isn't at the evidence level of the four ingredients on this list.

Does collagen in a cream increase my skin's collagen?

No. The collagen molecule is far too big to cross the epidermis: on the surface it hydrates and forms a film, but it doesn't turn into dermal collagen. What does work is prompting your skin to make it itself, which is the job of retinol, signal peptides and vitamin C.

What age should I start using anti-ageing ingredients?

Collagen starts declining from around 25-30, so that's the ideal window to begin with retinol as prevention, roughly 3-4 nights a week. Sunscreen, from always. And it's never too late: the landmark retinol trial improved fine wrinkles in people with an average age of 87.

Can I use retinol, vitamin C and sunscreen together?

Yes, but split across the day. Vitamin C in the morning alongside sunscreen, retinol at night. That's the textbook anti-ageing combo. What you shouldn't do is apply retinol during the day, or pair it the same night with exfoliating acids or benzoyl peroxide if you're a beginner.
